Carthagène des Indes fut le principal port colonial de la Nouvelle-Grenade et le siège d'un tribunal de l'Inquisition établi en 1610, l'un des plus actifs d'Amérique du Sud dans la persécution des judaïsants. Des conversos (marranes) s'y établirent malgré la surveillance inquisitoriale, participant au commerce atlantique tout en pratiquant certains rites juifs en secret. Plusieurs procès retentissants y eurent lieu au XVIIe siècle.
