Né à Lisbonne dans une famille juive portugaise, il s'installe en Virginie en 1745 où il exerce la médecine. Son père, Francisco Machado de Sequeira, médecin arrêté à Rio en 1706 et transféré à Lisbonne, se fit ensuite circoncire à Londres sous le nom d'Abraham de Sequeira Machado. John de Sequeyra a laissé des observations et écrits médicaux.
