Né de parents marranes au Portugal, médecin formé à Salamanque, il fut médecin en chef à Madrid sous le nom de Fernando. Ayant fui l'Inquisition, il revint au judaïsme à Venise (prenant le nom d'Isaac) puis s'établit à Vérone. Adversaire de la kabbale et du pseudo-messie Sabbataï Zevi, il écrivit une œuvre scientifique et une célèbre apologie du judaïsme.
