Né à Tivoli dans une famille juive italienne, Segrè découvre les éléments technétium et astatine puis l'antiproton, découverte pour laquelle il reçoit avec Owen Chamberlain le prix Nobel de physique 1959. Émigré aux États-Unis après les lois raciales italiennes de 1938, il participe au Projet Manhattan et devient professeur à Berkeley.
