Johannes Buxtorf II, fils du précédent, hébraïsant chrétien, naît à Bâle le 13 août 1599 et y meurt le 16 août 1664. Immatriculé à l'université de Bâle avant treize ans, il obtient sa maîtrise ès arts en décembre 1615, poursuit ses études à Heidelberg, puis assiste en 1618 au synode de Dordrecht; il succède à son père à la chaire d'hébreu de Bâle, refusant les offres de Groningue et de Leyde pour y rester jusqu'à sa mort. Marié quatre fois, il entretient, comme son père, une correspondance suivie avec de nombreux savants juifs — Jacob Roman de Constantinople, Salomon Gaï, Ménassé ben Israël, Jacob Abendana d'Amsterdam — et se livre au commerce des livres hébreux.
