Il est l'auteur de l'« Apologia Hebræorum », publiée à Strasbourg en 1559, dans laquelle il proteste contre le décret du pape Pie IV imposant aux Juifs des pays catholiques de porter un vêtement orange ou jaune pour les distinguer des chrétiens. Ascoli, comme Cinelli, qui louait l'ouvrage dans sa « Bibliotheca Volante », subit un long emprisonnement pour cette critique des autorités ecclésiastiques.
