L'un des premiers colons juifs de New York, alors New Amsterdam. Il fait partie d'un groupe de vingt-trois Juifs arrivés dans les Nouveaux-Pays-Bas en septembre 1654, apparemment après avoir quitté Bahia, au Brésil, reconquise par les Portugais sur la Compagnie hollandaise des Indes occidentales. Chaque membre du groupe s'étant porté garant individuellement des frais de traversée de tous, et les immigrants ne pouvant s'en acquitter entièrement, Ambrosius fut l'un des deux placés en détention civile par les autorités municipales ; il fut cependant relâché peu après et, comme ses compagnons, prospéra sur le sol américain sous pavillon hollandais.
