Né à Amsterdam dans une famille de Juifs portugais (le marchand de vins Isaac Haim Capadose et Esther Mendes da Costa), médecin formé à Leyde. Il se convertit au protestantisme en 1822 avec son ami Isaac da Costa et devint une figure du Réveil néerlandais. Adversaire déclaré de la vaccination antivariolique, il publia plusieurs écrits théologiques et médicaux.
