Disciple de Dov Ber, le Maguid de Mezritch. Il fonde en 1762 le centre hassidique de Karlin, pionnier du hassidisme en Lituanie (« Karliner » y devint synonyme de « hassid »). Réputé pour son ascèse personnelle et l'intensité mystique de sa prière ; racine des dynasties Karlin-Stolin et Koidanov. Le nom de famille Perlov vient de « Perl », prénom de sa mère.
